Determination of Malondialdehyde (MDA) Content in Rice Leaves

实验原理:水稻器官衰老或在非生物逆境条件下,膜脂发生过氧化作用,丙二醛(malondialdehyde)是其产物之一,通常利用它作为膜脂过氧化指标。MDA在高温及酸性条件下与TBA反应形成有色物,在532 nm波长处有最大吸收值,600 nm波长处有最小吸收值。
实验目的:检测水稻在非生物逆境胁迫前后膜脂的过氧化程度,检测水稻细胞膜对非生物逆境反应的强弱。

材料与试剂

  1. 冰盒
  2. 塑料袋
  3. 1.5 ml、10 ml离心管
  4. TCA (三氯乙酸)
  5. TBA (硫代巴比妥酸)
  6. 10% TCA (见溶液配方)
  7. 5% TCA (见溶液配方)
  8. 0.67% TBA (见溶液配方)

仪器设备

  1. 剪刀
  2. 研钵
  3. 100 ml容量瓶
  4. 天平
  5. 冷冻离心机
  6. 电磁炉
  7. 比色皿及分光光度计(DU640)

实验步骤

  1. 冰盒取样水稻叶片样品,置于封闭的塑料袋中,带回实验室立即测定。
  2. 天平称取0.5 g水稻叶片,剪刀剪碎后加5% TCA (见溶液配方) 5 ml,用预冷的研钵置于冰上研磨至匀浆状,倒入10 ml离心管中。
  3. 在3,000 rpm下冷冻(4 °C)离心10 min,取上清液2 ml至新的10 ml离心管中,向其中加入0.67% TBA (见溶液配方) 2 ml,混合后在100 °C沸水中煮30 min,室温自然冷却后取1.5 ml上清至新1.5 ml离心管中,室温12,000 rpm离心后取上清。
  4. 用分光光度计分别测定上清液在450 nm、532 nm和600 nm处的吸光度值。并按公式算出MDA浓度,再算出单位鲜重组织中的MDA含量(μmol/g)。
  5. 结果计算,按公式 C/μmol/L = 6.45 (A532-A600) - 0.56 A450
    式中A450、A532、A600分别代表450 nm、532 nm和600 nm波长下的吸光度值。

注意事项

  1. 检测生理指标的水稻组织质量应该一致,保证较好的实验结果。
  2. 检测植物生理指标所用试剂,最好现配现用。
  3. 用分光光度计测定吸光度时应注意分光光度计的测定范围。

溶液配方

  1. 10% TCA
    称取10 g TCA,使用ddH2O溶解,转移至100 ml容量瓶中定容
  2. 5% TCA
    将10%的TCA用ddH2O稀释一倍
  3. 0.67% TBA
    称取0.67g TBA用10%的TCA溶解,转移至100 ml容量瓶中用10% TCA定容
