RNA Extraction from Strawberry with Commercial Kit

摘要:本实验使用商业化的试剂盒进行,该试剂盒采用硅胶柱纯化方式,适合于从小于100 mg的植物组织中快速简单的提取总RNA。该试剂盒能彻底去除多糖类、酚类和酶抑制物等杂质污染。纯化的RNA可直接用于RT-PCR、Northern杂交和核酸保护等实验。提取过程无需酚氯仿抽提,也无需耗时的醇类沉淀。在30 min内可完成1个至多个样品的提取工作。使用硅胶柱纯化技术,大大减少了操作时间,减少RNase接触机会,因而该试剂盒对操作环境和操作者技能要求不高,即便是初学者也都能获得理想的结果。

材料与试剂

  1. 枪头
  2. RNase-free离心管 (1.5 ml,2 ml)
  3. 草莓果实
  4. E.Z.N.A® plant RNA kit试剂盒 (Omega Bio-tek, catalog number: R6827)
    1. Buffer RCL
    2. RWC
    3. RNA wash buffer I
    4. DEPC water
    5. HindBind RNA Spin column
    6. gDNA filter column
    7. 2 ml collection tube
  5. β-巯基乙醇
  6. Plantaid (博迈德, catalog number: RA107)
  7. 液氮
  8. 乙醇
  9. 10x Loading buffer (Takara, catalog number: 9157)

仪器设备

  1. 研磨用具
  2. 移液枪
  3. 振荡仪器
  4. 水浴锅
  5. 离心机
  6. 电泳仪
  7. 微量紫外分光光度计

实验步骤

  1. 液氮研磨果实样品并装入1.5 ml RNase-free离心管,每管样品量大约100 mg (具体根据果实发育不同时期含水量而异,如小绿果时期取样量小于 50 mg,成熟期果实取样量不少于300 mg)。
    注:本方法也可用于草莓叶片RNA的提取,每管样品量同小绿果,约50 mg。
  2. 加入500 μl buffer RCL (见溶液配方) (buffer RCL在使用前应加入β-巯基乙醇:每 1 ml buffer RCL加20 μl β-巯基乙醇),涡旋振荡混匀。
  3. 再向离心管中加入博迈得公司的plantaid 100 μl,再次涡旋震荡混匀。
  4. 55 °C水浴锅中裂解1-3 min后,可再次涡旋震荡。
  5. 室温14,000 x g离心5 min。
  6. 将上清转入gDNA filter column (装入2 ml collection tube),14,000 x g离心2 min (上清过多时可分2次转入)。
  7. 将滤过液转入新的2 ml RNase-free离心管中,加入等体积的buffer RCB,吸打混匀。
  8. 将混合液转入HindBind RNA Spin column (装入2 ml collection tube),10,000 x g离心1 min,弃掉滤过液,将column放回collection tube。
  9. 加400 μl RWC,10,000 x g离心1 min,弃掉滤过液和collection tube。
  10. 将column放入新的collection tube,加入500 μl RNA wash buffer I,10,000 x g离心1 min,弃掉滤过液。
  11. 重复上一步操作。
  12. 10,000 x g,2 min空离除去乙醇。
  13. 将HindBind RNA Spin column放入干净的1.5 ml RNase-free离心管,悬空滴入30 μl DEPC water至HindBind RNASpin column中的膜上,室温静置2 min,10,000 x g离心2 min,即得RNA溶液。
  14. RNA 质量的检测:取2-3 μl RNA溶液与loading buffer 混匀,用琼脂糖凝胶电泳检测RNA条带及是否有DNA污染。
  15. RNA浓度测定:取2 μl RNA 溶液用微量紫外分光光度计测定浓度并评估其质量。
