摘要:植物DNA的提取通常采用液氮研磨的方法破碎植物组织和细胞,通常需在抽提缓冲液中加入巯基乙醇以降低植物细胞匀浆中的多种氧化酶类对DNA抽提的影响。CTAB (十六烷基三甲基溴化铵) 是阳离子去污剂,可溶解细胞膜,它能与核酸形成复合物,在高盐溶液中可溶,当盐溶液浓度降低到一定程度时从溶液中析出,通过高速离心既可将CTAB与核酸的复合物与蛋白、多糖类物质分离,然后将CTAB与核酸的复合物沉淀重新溶解于高盐溶液中,再加入乙醇使核酸沉淀,再将沉淀DNA的溶于TE溶液中,即得草莓组织总DNA溶液。DNA的光谱吸收峰在260 nm处,蛋白的吸收峰在280 nm处,测定DNA含量时,通常计算OD260/OD280比值,该值为1.8-2.0时,DNA纯度较高,可进行后续PCR试验。
