实验目的:质构仪测量果实硬度精确且易操作,为果实硬度变化提供数据支撑。

材料与试剂

  1. 草莓果实

仪器设备

  1. 计算机
  2. 质构仪 (Stable micro systems, UK)

实验步骤

  1. 打开电脑,桌面的质构仪软件,然后打开质构仪开关
  2. 设置参数 (图1)
    T. A. Setting菜单栏里的library里有7个模式,选择1. Return to start,(一般选这个,OK);进入:
    Test Modle:compression;
    Pre-test Speed:1 mm/sec;
    Test Speed:1 mm/sec;
    Poet-Test Speed:10 mm/sec;
    Target Mode:Strain;
    Strain:20-30%;
    Trigger Type:Force,5.0 g。

  3. Test Configuration (图2) 
    1)
    Archive Information:选择存储路径和样品名称命名;
    2)
    Probe Selection:二倍体草莓:Unknown;八倍体草莓:P/2;
    3)
    Parameters:7 Strain Height,Auto;8 Stress Area,Option,优选小的,一般选探针Probe;
    4)
    Data Acquisition:Acquisition Rate,10,若较软,选较小值,较硬则选偏大的值;
    5)
    OK。

  4. 力量校对 (图3)
    点击力量校对按钮;下一步;放置校对金属块;下一步;1000.0正常。

  5. 高度校对 (图4)
    让探针靠近平台校对。
    注:力量校对和高度校对很重要,否则易损坏样品及探针。

  6. 抬高探针,放置待测样品,尽量避开种子位置,点击测试按钮Run a test。
    注:若探针失控,一直没减速,需按紧急STOP按钮,否则探针被挤压在平台,损坏探针。
  7. 存储数据
    1)
    点击保存按钮,输出;
    2)
    右键,DATA,全选,复制粘贴至文本,再把数据从文本中粘贴到excel。
  8. 每个处理或者时期测至少三个果实,每个果实至少测两个位置 (Video 1)。

结果与分析

由表1得随时间的推移,红颜草莓的果实硬度先增加后降低再增加再降低,可见红颜草莓的果实硬度是由于果实组织的不同而发生改变。
