Identification of Citrus Pollen Vitality

实验原理及目的:在育种过程中,花粉的质量和数量直接影响育种的成败,因此花粉活力的鉴定显得尤为重要。花粉的活力可直接影响产量,有活力的花粉会被醋酸洋红和氯化三苯四氮唑 (TTC) 染成红色,被碘-碘化钾 (I-IK) 染成蓝紫色。

材料与试剂

  1. 载玻片

  12. 1%醋酸洋红 (见溶液配方)

  3. 万能显微镜 (Olympus, model: BX61)

实验步骤

  1. 醋酸洋红染色法
    1.1
    在载玻片上用吸管滴一滴1%醋酸洋红,用毛笔轻蘸烘干的柑橘花粉,轻轻洒在醋酸洋红上。
    1.2
    盖上盖玻片,盖时用镊子夹住盖玻片一角,使之倾斜与载玻片呈30-45°,缓慢放下盖玻片,防止气泡产生,并用滤纸吸干多余的醋酸洋红。
    注:盖盖玻片时,若产生气泡会影响显微镜观察和拍照,多余的醋酸洋红会使花粉移动,也不利于显微镜观察和拍照。

    染色一分钟后,于万能显微镜下观察拍照。每个品种3-4个重复,每个重复选取多个视野拍照,保证总花粉数多于500个。用软件Image J统计花粉染色数量与总的花粉数量,计算比值即为花粉活力。
  2. I-IK染色法:此方法适用于不含淀粉的不育花粉,只染成蓝紫色表明花粉发育良好、活性强,呈黄褐色为发育不良的花粉。

    取1 g的碘化钾溶于10 ml蒸馏水中,然后加入0.5 g碘,待全部溶解后,再加蒸馏水至200 ml,配好试剂后,置于棕色瓶中备用。

    将一滴I-IK溶液滴在载玻片上,用毛笔均匀撒上花粉,盖盖玻片,放置1-2 min进行统计,方法同醋酸洋红染色法。
  3. TTC染色法:具有生命力的生物体在脱氧酶催化的氧化还原反应中,把氧转移给TTC,使其生成红色的苯甲潜 (TTFC),因此,凡染为红色的花粉粒是具有活力 的,呈淡红色的活力较低,无色的是没有生命力的。
    3.1
    用镊子在载玻片上放10粒花药,滴蒸馏水1-2滴,用镊子夹出花药,清除花药壁后,滴0.5% TTC 1-2滴,盖上盖玻片,于黑暗中染色30 min。
    3.2
    每个处理做1-3个重复,染色完毕后显微镜下观察着色情况。每个重复选取6个视野拍照,进行统计,方法同醋酸洋红染色法。

溶液配制

  1. 1%醋酸洋红
    洋红1 g,45%醋酸100 ml,煮沸2 h左右,并不断补充蒸馏水,保证其总体积不变,之后冷却过滤,加入4%铁明矾 (硫酸铁铵) 溶液1-2滴 (不能加多,否则会发生沉淀),放入棕色瓶中常温保存备用
