实验原理:低温、干燥环境下水稻种子处于较好休眠状态,不利于虫子及杂菌生长,保持种子使用时的活力。
实验目的:方便种子取放及较高的发芽率。

材料与试剂

  1. 水稻种子

仪器设备

  1. -20 °C冰箱
  2. 低湿柜 (明日百傲,HZM-400)
  3. 抽真空仪 (MAGIC SEAL/美吉斯PR4257) 及真空袋 (尼龙,5cm*10cm)

实验步骤

  1. 种子收获,水稻种子进入黄熟期便可以收获,成熟种子长期不收易长霉染菌或发芽,不利于种子的贮存。
  2. 种子干燥,黄熟期种子收获后,晴朗天气下暴晒一周时间即可脱粒、装袋、编号。
  3. 低湿柜存放种子,耗电低,柜内湿度10%-15%,但由于低湿柜不能控温,种子挂藏室内温度高,建议夏天7月-9月空调降温。此外,低湿柜较大,为方便取放,建议定制低湿柜用抽屉按姓名编号存放。
  4. 冰箱存放种子,耗电大,种子干燥后入纸袋,放于真空塑料袋中,抽真空后置于-20 °C冰箱。但经常取放,冻融不利于种子保存。
  5. 长期不动种子即备用重要种子推荐使用超低湿柜如发表文章材料,柜内湿度低于10%。

注意事项

水稻种子发芽及水稻幼苗生长实验,实验所需种子需要同期收获,并且在同样保存条件下的种子(图1)。
