实验原理:真核细胞mRNA最显著特征是具有5’–端帽子结构和3'–端的polyA尾巴。mRNA的polyA与oligo(dT)纤维素柱在高盐下亲和吸附。在低盐下吸附解除,因此可利用该原理分离mRNA,再用有机溶剂沉淀可得纯化mRNA。 实验目的:从水稻组织中分离纯化mRNA,可以用于逆转录,RACE和文库构建等后续实验。
