果蝇幼虫的饥饿处理实验

摘要:脂肪是体内储存能量的重要物质,在饥饿条件下,可能会调动脂肪代谢。为了研究哪些物质参与脂肪代谢,可以通过饥饿处理刺激基因表达进行进一步研究 (Tian等,2011)。本实验方法阐述了如何进行果蝇幼虫的饥饿处理实验。

材料与试剂

  1. 培养皿,直径60 mm (海门市昊宏实验器材经营部)
  2. 培养管
  3. 滤纸
  4. 果蝇成虫 (100~120只,雌雄比为5:4)
  5. 蒸馏水
  6. 酵母

仪器设备

  1. HP400S生化培养箱 (武汉瑞华仪器设备有限责任公司)

实验步骤

  1. 将刚发育完成的成虫 (100~120只,雌雄比为5:4) 在培养管中 (加酵母) 培养2天。
  2. 每隔两小时收一次卵 (保证果蝇的发育基本在同一时期)。
  3. 收集到的卵在25 °C培养箱中培养3.5~4天。
  4. 取培养管表面一半的幼虫,用蒸馏水将幼虫表面食物洗干净并用滤纸吸干幼虫表面残留的蒸馏水 (培养管中另一半幼虫作为对照组)。
  5. 将洗干净的幼虫放入垫好滤纸 (事先浸润过蒸馏水) 的培养皿中,每个培养皿中放入50~60只幼虫。
  6. 在25 °C培养箱中进行饥饿培养,分别在12 h、24 h、36 h取饥饿处理过的幼虫,进行下一步实验。
