实验原理:农杆菌是普遍存在于土壤中的一种革兰氏阴性细菌,它能在一定条件下趋化性地感染植物的受伤部位,并诱导产生冠瘿瘤或发状根。根癌农杆菌和发根农杆菌细胞中分别含有Ti质粒和Ri质粒,它们都含有一段可转移的DNA(T-DNA),农杆菌通过侵染植物伤口,可以将T-DNA插入到植物基因组中,并且稳定的遗传给后代。本实验是把目的基因装载到经过改造的Ti质粒的T-DNA区,借助农杆菌的感染实现外源基因向水稻愈伤的转移与整合,然后通过组织培养技术,再生出转化植株。 实验目的:通过农杆菌介导的遗传转化把目的基因导入到籼稻品种中(MH63,ZS97,9311等)。
