Determination of ACC Content in Rice Tissues

实验原理:ACC(1-氨基环丙烷-1-羧酸)是乙烯合成的前体,植株完整生长发育期难以检测气态乙烯的含量,通常以ACC含量指示植物体内乙烯的相对含量。本实验用气相色谱方法检测反应后生成乙烯的量。
实验目的:通过检测水稻组织中ACC的含量,间接反应水稻组织中乙烯的含量。

材料与试剂

  1. 1.5 ml离心管
  2. 各种型号的枪头

  3. 液氮
  4. 氮气
  5. 甲醇
  6. D2ACC
  7. NaOCl
  8. NaOH
  9. 10 mM HgCl2
  10. 5% NaOCl:饱和NaOH = 2:1 (v/v) (见溶液配方)
  11. 抽提液 (见溶液配方)

仪器设备

  1. 离心机
  2. 天平
  3. 玻璃瓶
  4. 气相色谱 (岛津)
  5. 脱色摇床
  6. 氮吹仪 (Dry N-EVAPTM111, Organomation Associates Inc. USA)
  7. -70 °C冰箱

实验步骤

  1. 取样
    液氮取水稻叶片,每份样品三个生物学重复,每份样品大约0.1 g鲜重,-70 °C保存直到抽提,避免反复冻融。
  2. 抽提
    2.1
    液氮研磨,磨碎;取约0.1 g于1.5 ml管中,加750 μl抽提液,颠倒混匀,置冰上,避光。

    4 °C、脱色摇床300 rpm避光抽提4小时;4 °C、13,000 rpm离心10分钟,吸上清于一新离心管中。
    2.3
    在沉淀中加入450 μl抽提液,4 °C、脱色摇床300 rpm避光抽提4小时,4 °C、13,000 rpm离心10分钟,吸上清,合并两次上清于玻璃瓶中。
    2.4
    氮气吹干,加入390 μl纯水定容(检测ACC一般不用纯化)。

    在冰上加入10 μl 10 mM HgCl2
    2.6
    在冰上加入100 μl 5% NaOCl:饱和NaOH = 2:1(v/v),放冰上3分钟
    2.7
    气相色谱火焰检测器检测。

注意事项

  1. 抽提液配成mixture,-20 °C保存长期使用。
  2. 所用试剂5% NaOCl:饱和NaOH = 2:1 (v/v) 及10 mM HgCl应4 °C保存,使用前应置于冰上。
  3. 所用试剂除D2ACC购置于OlChemIm,其余均为国药试剂。
  4. 抽提过程注意避光和低温。

溶液配方

  1. 抽提液
    80%甲醇
    加5 ng/μl D2ACC (H2标记的ACC做内标)
  2. 5% NaOCl:饱和NaOH = 2:1 (v/v)
    注:氢氧化钠饱和溶液:称取120 g氢氧化钠,加100 ml水,振摇使之溶解成饱和溶液,冷却后置于聚乙烯塑料瓶中,密塞,放置数日,澄清后备用。
