Dear Dr.Wang,
When I transformed the TevCas9 vector into the Phaeodactylum tricornutum, I found there were multiple cuts of the target gene of the single clone grown on the zeocin plate .
I only design the gRNA of Cas9 and ignore the TEV site. I used gene gunning to bombard the vector into the Phaeodactylum tricornutum.
I would appreciate it if you could help me solve this problem.
