Skin penetrating IL-RNA complexes
IL synthesis
1. The cation, choline bicarbonate and the anion (for eg. geranic acid) was mixed at a 1:2 ratio to prepare IL following salt metathesis reaction.
2. Geranic acid (3.696 mol) was weighed in a round bottom flask equipped with a stirrer and placed into a water/oil bath.
3. Choline bicarbonate 80% solution (1.848 mol) was added dropwise to the flask. The mixture was reacted for 24h at 40°C.
4. The resulting IL solution was dried using a rotary evaporator at 20 mbar at 60 °C for 2 h.
5. The residual water was removed in a vacuum oven at 60 °C for 48 h and characterized.
Preparation of IL-RNA complex
1. ILs were mixed with RNA (100µM) at a volumetric ratio of 1:1 and incubated for 30mins at RT.
2. The RNA-IL solutions (1ml) were dialyzed against 10mM sodium phosphate buffer for 72h using Dialysis Cassettes (10,000 MWCO, Invitrogen).
3. The concentration and stability of RNA were determined.
