Cell Culture and Drug Treatment

The human NSCLC cell lines A549, SK-MES-1, H460, and SPCA1 were obtained from the Cell Bank of Type Culture Collection of Chinese Academy of Sciences (Shanghai, China) and were cultured in Dulbecco’s modified Eagle’s medium (DMEM, Gibco, Waltham, MA) supplemented with 1% penicillinstreptomycin and 10% Fetal Bovine Serum（Gibco, Waltham, MA）at 37°C with 5% CO2and 21% O2.
For cell proliferation assays, 5x103 NSCLC cells/well were seed into 96-well plate. After 24 hrs, cell treated with indicated concentration of DPT, control wells added DMSO as 0 point. Harvest cells for detection at 12, 24, and 48 hr points. 
For RNA or protein harvest, 2x105 cells NSCLC cells/well were seed into 6-well plate. After 24 hrs, cell treated with indicated concentration of DPT, control wells added DMSO as 0 point. Harvest cells for further detection after 24 hr treatment.
