Thioflavin S (TS) staining
TS is a dye commonly used to label the dense core of senile plaques for evaluating Aβ burden. TS (T1892-25G, Sigma-Aldrich) was first dissoved in 50% ethanol (ethanol:ddH2O, 1:1) to make a final concentration of 2% (M/V) TS stock solution and and keep it in dark at 4℃. Before use, the TS working solution was prepared by dilluting the stock solution in 50% ethanol to a final concentration of 0.002%. For staining, brain sections were stained with the TS working solution in the dark for 8 min at room temperature, followed by two washes with 50% ethanol and three washes with PBS. Afterwards, brain sections were mounted for imaging or processed for further immunofluorescence co-staining with other antibodies.
