Immunofluorescence Protocol

Quintana lab

1. Rinse sections of antifreeze solution with PBS pH 7.4........................…………………………................. 3x5’
2. Block sections with PBS 0.2% Triton X-100 + 10% normal donkey serum (NDS) (9ml +1ml) ......…....... 1h at RT
3. Incubate with primary antibody in PBS + 0.2% Triton X-100 + 1% NDS (9.9ml +0.1ml)...........…............ ON at 4ºC
4. Wash with PBS 0.2 Triton X-100.....................…………………………………………………….................. 3x5’
5. Incubate with secondary antibody PBS + 0.2% Triton X-100 + 1% NDS (99ml +1ml)...................…....... 1h at RT
6. Wash with PBS 0.2% Triton X-100...........…………………………………………………. ........................…1x5’
7. Wash with PBS.........................…………………………………………………………………………..........…1x5’
8. Wash with PB…................................………………………………………………………………………........ 1x5’
9. Mount stained sections onto slides in PB
10. Coverslip with Fluoromount G with DAPI
11. Seal the edge with nail polish and leave to dry 2h before visualising
12. Store at 4ºC protected from light
13. Analyse within 1-2 weeks

Buffers:

PBS

1 tablet + 1000ml H2O

PBS 0.2% Triton

1000ml PBS + 2ml Triton X-100                                             500ml PBS + 1ml Triton

Incubation volumes:

2ml/well 1ml/flask

0.5ml/eppendorf 1.5

Normal Donkey Serum (NDS) -20ºC/box1 1º antibodies: -20ºC/box6

2º antibodies: +4ºC, Chicken 488 -20ºC/box6

PBS tablet – Millipore 524650-1EA
