The protocol is described in the following publication: Cohen, M. A. et al. Human neural crest cells contribute to coat pigmentation in interspecies chimeras after in utero injection into mouse embryos. Proc Natl Acad Sci USA 113, 570–1575 (2016).
“Microinjections to E8.5 Embryos. Pregnant females at E8.5 were anesthetized via an i.p. injection of avertin. Laparotomy was per formed and the uterus was exposed and held with forceps during the injection of each embryo. NCCs were drawn into a glass mi cropipette, and the tip of the glass micropipette was inserted into the distal third of the decidual swelling. Approximately 2–5 × 103 cells (suspended in 0.2–0.8 μL of cell culture medium) were injected per embryo.”
