Open the image using the Fiji image analysis software (http://fiji.sc).
Launch the ROI Manager and enclose four epidermal cells above and below the boundary between the meristematic zone (MZ) and the transition zone (TZ) using the Rectangular tool. Note that the measurement is conduced only in the hair cell layer, and that the boundary between the MZ and the TZ is defined as the point where the epidermal cell length in the TZ is twice of that in the MZ.
Register selected regions of interest (ROIs) in the ROI Manager.
Measure the GFP fluorescence intensity for registered ROIs.
