Incubate 37ºC for 30 mins, heat inactivate 65 ºC.
Digestion of backbone
Digest backbone pPBhCMV_1-miR(BsgI)-pA-3 (1µg) with Bsg1 supplied with SAM.
Gel purified. Following this, the annealed primers and purified digested vector were ligated at a 15:1 insert to backbone molar ratio using Instant Sticky-end Ligase Master Mix (M0370S, NEB).
