To obtain 100k intestinal stem cells, I dissected about 120 flies. Depending on the state of your flies this may vary. 

For the ATAC protocol, I followed the steps in Buenrostro et al., 2015, which describes the protocol in great detail (doi: 10.1002/0471142727.mb2129s109).
