Corneoscleral tissue is rubbed with a cotton bud on the posterior side to remove endothelial cells, scraped on the anterior side to remove epithelial cells and washed in PBS. The corneal/limbal region is excised and cut into small pieces prior to enzymatic treatment for limbal cell isolation.
