Do we really use "lysis buffer" at step A10 and not "RBC lysis solution"?

The required volume of 5 ml seems strange when only 4 ml of of lysis buffer is prepared per sample, and it is all used in section C for nuclei extraction. Also section A does not seem like the section where we would want to lyse cells.
