I am sorry that I did not receive any E-mail notification for your question. Sorry for the delay. Thank you very much for your positive comment. For your technical problem, I suggest to increase the incubation time of permeability with 0.5% Triton-100, you can include 0.5% Triton-100 in the blocking buffer for 1 hr incubation or double check the primary Pax7 antibody, whether it is from DSHB and secondary antibody it should be IgG1a specific not H+L.
