Hi, very nice protocol.
I wonder which mouse strain did you use in this study?
Do you think a naive C57BL/6 mouse has enough mature osteoclasts to isolate?

Thanks for the questions.
As for the question on mouse strain, we have tested this protocol in C57BL/6J mice. I think it could be applicable for the other mouse strains.
As for the question on mature osteoclast number isolated from one mouse, it will depend on the cell yield during the protocol (pls refer to the trouble shooting part in 'Notes'), and the bone remodeling status of the mouse, e.g. with OVX. You may consider to pool 2-3 mice together along with LS column to get a higher cell yield,
