For the colour observation step, was there a certain OD600 that should be obtained before harvesting the cells? And did you confirm your YTK12 transformants via PCR or just by growth on the different medium? Thanks!

1. In our assay, we did not measure OD600, and I think it is a good idea. Because it is possible to observe different degree level if red when you test different signal peptides.
2. For the first, we select the colonies that can grow on the minus Trp medium, and then the colonies will be confirmed via PCR. The positive colonies are selected for next steps.
