Dear Tina,
The number of discs, the volume of the well, and such are by no means appropriate in different cases. If your conductivity meter requires a large volume, you would need to put a higher amount of tissue - the sensitivity of the measurement depends on tissue amount/water volume. On the other hand, if you use a larger amount of tissue, the response of a higher number of cells get averaged, which may result in an underestimate of SD in the cell response. You just need to decide the scale of measurement considering the practicality and the potential trade-offs in different experimental system.
