Is it still important adding sucrose if the embedded tissue is only applied for vibratome slicing ? Thank you

The original protocol was written for a freezing microtome and I have never attempted to change the solutions when we found the vibratome more reliable (the freezing microtome shredded sections from time to time due to knife chatter). As a reference, you can check out Bahr et al, 1957. There is evidence that high concentrations of sucrose will prevent swelling of the tissue post-fixation. 10.1016/0014-4827(57)90148-9
