We usually stain dystrophin on muscle cross-section just using typically fixation with 4% PFA for 10min. The staining is always good. For whole muscle tissue, it can be fixed for 2-4 hours dependent on the muscle size, and processed with glycine for 2hours, PBS wash 0.5h following dehydrate with 15% and 30% sucrose overnight. The staining is also good no matter is in embryo or adult muscle tissue.
