I assume you have labeled the peaks correctly, although we have not specifically tested to be sure those peaks represent the 40S, 60S and 80S. We also have not calculated changes in these subunits, although I would think you could calculate the area under the curve for each peak and represent as a ratio or percentage of the total area under the curve to account for any differences in sample loading. To me it looks as though your KO samples may be slightly underloaded compared to the WT samples as all the peaks seem to be diminished compared to the WT.
