Thanks for all your hard work developing this protocol. I was wondering if you have tried (or have throughts about) using a 10x stock of ACSF (stored at 4 C) rather than preparing it fresh each time.

Hi Damian,

Thank you for your questions.
I have used 10x stocks in the past without any issues.
But I have then developed a habit of making fresh aCSF daily which gives more flexibility when one does studies that require changing the composition a slightly bit (potassium to modulate excitability, glucose to remain closer to physiological concentrations, etc.) Overall this also saves time from the thawing process.
A 10x stock also exposes you to possible chronic 'issues' with your aCSF if there was any sort of problem with the powders or the water source when you made the stock solution.
It comes down to personal preferences. I teach my students to make it fresh every day. But I have no problem if they prepare a 10x stock.

I usually prepare 10X stocks and use them for 7-10 days without any issue
