Can I spin the peripheral blood sample to remove plasma( for other studies ) and reconstitute the rest of the sample with DEPC-PBS , before layering on to Ficoll?Should I be adding Human serum to PBS before doing the same?

Yes, you could separate the plasma. and dilute the cells with PBS 1:1. No need to add human serum to PBS before layering.
