I have tried with 2% raffinose, all the clones were growing easily. Should I reduce the concentration of raffinose in YPRAA medium?
At 2% I am not getting any positive selection. mentioned here.

Actually, antimycin A is indispensible in YPRAA, if you omit antimycin from YPRAA, even YTK12 and YTK12 transformed with empty vector can grow well.
