I would like to know how long it will take for E1 buffer and E2 buffer to crack cells respectively after the addition of E1 buffer and E2 buffer in steps 4 and 13 to centrifuge? Thank you for your answe.

The dissolution of respective membrane is fast. By pipetting up and down few times and by the time you process all your samples, the cells will be lysed before you start the centrifugation.
