To prepare SeeDB2G, dissolve 10g Histodenz in 7.5 mL Tris-EDTA (~57.1% w/w) as described in Recipes. The refractive index will be exactly the same as that of Omnipaque350 (1.4662).

In most applications, there will be no need for further refinement. Please note, however, that the immersion liquid for some glycerol lenses may have a slightly lower refractive index. For example, 92% w/w Omnipaque350 matches Type G immersion liquid.

As for Triton X-100, we recommend 0.1-0.5% to minimize morphological damages. 2% is too high for Triton X-100.

To further minimize morphological damages, we also suggest more gradual clearing with intermediate concentration steps.
