We generally grow Pseudomonas aeruginosa in 250ml flasks with shaking, using 25-50 ml of growth medium. For biofilms, we used a tube reactor system, and collected biofilms as a paste into microfuge tubes. What is important is that for c-di-GMP extraction, similar ODs are being used. For all samples regardless of growth conditions, we made use of 1 ml adjusted to an OD600 = 1.8
