I have successes with as few as 20-30 adult worms, using the standard Trizol extraction method as mentioned above.Although for a sample size that is fewer than 50 (L4-adult) worms, there are extra steps that need to pay special attention. For example, at step 4, it had better to vortex (violently) on the tabletop (Fisher, cat.log #12-812) to ensure complete dissociation of the worm tissues; at step 10, it helps to precipitate small amount of RNA by adding 5-10ug glycogen (invitrogen cat #1466580) as a co-precipitant; at step 10g, ice-cold EtOH is also useful to precipitate RNA.
However, if you start with a large number of materials (>100 worms), just follow the protocol will usually yield about 10-20ug total RNA.
