protocol Protocol: Assessment and Quantification of Foam Cells and Lipid Droplet–Accumulating Microglia in Mouse Brain Tissue Using BODIPY Staining

Thank you for the detailed protocol. Regarding the staining step where sections are incubated in a solution of 1:25 BODIPY 492/515 in DMSO for 30 minutes, could the specific preparation steps be clarified? The supplier recommends a 1 mg/mL stock solution. What is the final concentration Read more Read less more less

Thank you for your question. To prepare the stock solution, follow the supplier's instructions to make a 1 mg/mL solution in DMSO. This stock solution can be stored at -20°C for up to one year. I recommend aliquoting it into smaller volumes (e.g., 1 mL) to avoid repeated freeze-thaw cycles.

For the working solution, dilute the stock solution further with DMSO to achieve a 1:25 dilution as specified in the protocol. For example, to prepare 1 mL of working solution for the staining step, combine 40 µL of the stock solution with 960 µL of DMSO. I hope this addresses your question clearly.
