What must be the reason of not colour change in the sample even after addition of Na2Co3?

Dear Shreya,

if you have just tried all the reagent through making the standard curve, the only reason why you don't see a color formation should be the lack of any beta-glucodidase activity.

Sometimes impurities in water or reagents can interfere with the colouring, but if you get the standard curve right, then it is as I said above. You could also test a positive control using a collection strain (e. g. L. plantarum DSM 20205) with proven enzyme activity. I hope you will solve the problem. Best regards.

Thanks a lot for answering my question but the thing is when I increased the concentration of enzyme like- 1ML then I can see the colour change but if I dilute the enzyme to 1/10 dilution then there is no colour change in the sample
