I was wondering at what wavelength DPBA was excited. also, did you encounter autofluorescence in that specific channel?

If there was autofluorescence, what microscope settings were used to distinguish auto fluorescent signal from DPBA signal?

Hi Hana,

Based on my own experience, I did not really observe the fluorescent signals (or the signal was very low) when I used the attenuated flavanols mutants such as the Arabidopsis ttg1 line. Thus, it is better to include the controls in your experiments such as non-treated samples, etc.

Regarding the excitation and emission wavelengths,

I am sorry for the missing info here.

The excitation is 488 (Argon) (for Kaempferol) and 561 (Hene) (for quercetin).

Let me know if you have any more questions.

Best wishes,

Nguyen
