How we can calculate the alginate beads drug entrapment efficiency by indirect method?

Please give me the reference. Should we crush the beads then put it in a phosphate buffer solution 7.4 for 30 minutes and centrifuge for 60 minutes and then separate the supernant and then measure its absorbance? We put this formula:
