Could you please share more specifically how the CM4620 mix was prepared for the gavages? Thank you.

We followed the prep from CalciMedica that they gave us when we received CM4620. In order to do in vivo work in mice, you need spray-dried dispersion form of the drug.

We made 0.5% methyl cellulose and 1% Tween 80 in water the night before. The next morning we took out the vehicle and the spray dried version of CM4620 and allowed them to come to room temperature. We then made the mixture for the day (40 mg into 9.6 ml of vehicle) and used an electric homogenizer for 5 mins and then performed oral gavage.

Thank you for the clarification on the preparation of the CM4620 above. Could you please clarify what you mean by electric homogenizer? Is it an ultrasonic homogenizer (sonicator) or a mechanical homogenizer?

Thank you very much for your quick response. This preparation, however, provides a dose over 40 mg/kg given the 10 ml/kg gavage preparation. In the article, it's stated that the dose was 10 mg/kg. Could you please comment on that? Thank you very much again.
