Can this be scaled up to a cell pellet as opposed to a colony?

protocol Protocol: A Quick DNA Extraction Method for High Throughput Screening in Gram-positive Bacteria

I would like to grow out R. Gnavus in 5 mL liquid culture and do a quick extraction of genomic DNA from a cell pellet. Would this work? Would I have to increase the time of sonication?

Sure, in our system, we have successfully extracted genomic DNA from 1 mL liquid cultures of various Gram-positive bacteria. Using a cell pellet facilitates the screening of transposon mutants. For R. Gnavus, of course you can try our protocol and I recommend checking the instructions of commercial kits to see if any special enzymes are required to break up the cell wall. If such kits are not available, consulting the existing literature or experts in the field can provide valuable insights for optimizing the DNA extraction protocol. Good luck with your research!
