In absence of a fraction collector, fractions from an ultracentrifuged sucrose column can be collected manually, but has to be done very carefully so as not to disturb the column and destroy the gradient. Either it can be done from the top by taking out equal volumes very slowly using a pipette. Or a hole can be made at the bottom of the tube by using a heated needle, and then collecting equal number of drops for each fraction. Both of these have been used in labs for long but requires some skill and careful handling.
