How to keep the tissue away from shrinking during imaging?

protocol Protocol: Colocalization Analysis for Cryosectioned and Immunostained Tissue Samples with or without Label Retention Expansion Microscopy (LR-ExM) by JACoP

When the tissue is placed on the microscope stage, which detached from the water, it shrinks during imaging. This greatly affects the expansion factor. How do I keep it from drying out during imaging?

I would suggest to keep the water in the well till the start of imaging. After imaging trial done under the confocal, add water back to the well. Or you can add a little bit 0.8% low melting agarose at the edge of the gel to keep the moisture and position of the sample in the gel on the glass bottom plate.
