L929 cell-conditioned media is commonly used to differentiate bone marrow-derived monocytes (BMDMs) into macrophages in vitro. The conditioned media from L929 cells contains colony-stimulating factors that promote the differentiation of monocytes into macrophages. The inducted have already undergone cell differentiation and therefore you don't need to add additional L929 cell-conditioned media in the following experiments.
