Under acidic conditions, proline can react with ninhydrin to produce a stable red product with a maximum absorption peak of 520 nm. 


The absorption rate at 520 nm is proportional to the concentration of red product in the solution after toluene extraction.  Therefore, the content of proline in the sample could be determined by standard curves or computed by regression equation.
