Hello dear Vikas Singh!
Very nice protocol but I still have some small questions:
Is there a reason the amount of EDTA disodium is 0.16 g instead of 0.186 (0.005 M in 100 ml) and K2HPO4 0.85 instead of 0.87 (closer to 0.1 M)?

Hi Leonhard
For EDTA-disodium dihydrate 0.186 g is correct and K2HPO4, 0.87 gm is correct.
Thank you for pointing out the mistake.
For OPT solution, it is easier to weigh 10 mg instead of 1 mg. Therefore, we prefer to make 10 mg/ml stock rather than 1 mg/ml.
I hope this will answer your queries.
Thank you
Vikas
