This method can be used to study transcription factor binding, e.g. shown with RB:

To study transcription factor activation by this method, the protein would have to be activated by being bound more to chromatin. Alternatively, if the transcription factor is phosphorylated after activation, this could also be measured by this method using a phosphospecific antibody. However, every new antibody used would require validation.
Furthermore, if a transcription factor globally activates transcription, then this might be expected to cause higher pRNAPII S5 and S2 levels. E.g. https://www.ncbi.nlm.nih.gov/pmc/articles/PMC6318124/
