What I did was RNA interference experiment, and only 3 groups were set, namely blank control group, negative control group (NC), and experimental group. However, after each real-time quantitative PCR, it was found that the blank control group was very different from the negative control Read more Read less more less

Hello!

There will be some differences between the blank control and NC group. Because the transfection of small RNA will certainly have a certain impact on the cell itself, and then affect the expression of some factors, which belongs to the influence of experimental operation. But in your experiment, the ratio of blank control group to negative control group was 0.6:1, is it before or after the reference? If there is no reference correction, then the comparison is meaningless. If it is after reference correction, then your blank group is different from the NC group in question. I'm guessing there might be something wrong with your NC clip.

Good luck!
