How to deal with ELISA blank hole?

My experiment was to measure interleukin 4. I would like to ask you that WHEN I made ELISA, I set two OD values of blank hole as 0.072 and 0.034 respectively. Hello!

Your question depends on how you set it up. Because the blank value is subtracted from the whole board (standard product, quality control and sample to be tested are subtracted at the same time), it is not just subtracted from standard product alone. If you want to subtract the average of blank holes, you can set it directly in the software of the enzyme standard instrument.

If you don't subtract the whitespace, you don't need the whitespace when you're doing the curve, it's just a reference. In fact, subtracting or not subtracting blank values has no effect on your curve or sample.
