when I get my picture of cell fluorescence in immunohistochemistry experiments. And I would like to measure the intensity of green fluorescence in the nucleus using ImageJ. Is there any smart way to do this?

ImageJ can be a very convenient software for processing image information. For fluorescence determination, you can
1. Loading the image to ImageJ
2. Convert image format: RGB →8 bit
3. Choose a threshold in your channel
4. Set a measurement: Area (the Area of the selected Area); Mean (Mean gray value of the selected area of the box); IntDen (Integrated Density) (total fluorescence intensity in the selected area);
5. Analysis
